The 'R Line' is just a trim level, VW's equivalent of 'Sport'.
It will have large alloy wheels (probably 18 or 19 inch) which will mean tyres are more expensive than on the standard model, 'sports' seats with larger side bolsters to stop you sliding around in corners, possibly some fancy headlights and probably a fair amount of fake carbon fibre or fake aluminium trim inside instead of the fake wood used on the 'luxury' version or grey plastic in the normal version.
For a 6 year old car, none of these particularly affect its value or desirability. It would be worth checking the coat of replacement tyres for it though as it may be higher than you'd expect.
