I want to buy off eBay a rare collection DVD but it is only one that I can find is in American. I have never bought anything outside of UK before, I am not perfect in maths but how to work out the import charge?

The DVD is cost $50.00 with $12.00 postage to UK from Canada?

If you are buying from a private individual ask him to mark the Canadian customs declaration as "gift" and the value as being the ORIGINAL cost of the DVD.
This way you should avoid all import charges.

It all depends on the value of the item in GBP, there is no customs duty on items under £135. You will pay VAT on gifts worth more than £34 or other goods worth more than £15. If your item is chargeable there will also be a handling charge made by the company who delivers it in the UK. Royal Mail charge an £8 handling/admin fee but, others might charge more.

Bit late but if VAT is charged, my understanding is that it is calculated on the basis of the value of the goods and the postage charge. I presume you are able to play N. American DVDs.
